    Sorghum

    Sorghum (Sorghum bicolor [L]. Moench) is a crop indigenous to Africa, where it appears to have been domesticated in Ethiopia about 5000 years ago. It is now widely cultivated in dry areas of Africa, Asia, the America, Europea and Australia between latitudes of up to 50 degree Nothe in North America and Russia and 40 degree South in Argentine. Sweet sorghum is a variety closely related to grain sorghum; it differs mainly in that its stalks are taller and juicier with higher sugar content than the grain sroghum type. Sorghum is the firft most important cereal in the world after wheat, rice maize and barley. In Africa it comes second after maize in terms of production. Sorghum is well adapted to tropical climates with several traits making it a drought-tolerant crop that survices under adverse climatic conditions, and thus is often relegated to poor soils and low-input management. It is extensively grown under rainfed conditins for grain and forage production. High productin may be achieved when sufficient water and nutrients are applied especially at critical stages of crop growth

    Functional homogeneous zones (fHZs) in viticultural zoning procedure: an Italian case study on Aglianico vine

    Abstract. This paper aims to test a new physically oriented approach to viticulture zoning at farm scale that is strongly rooted in hydropedology and aims to achieve a better use of environmental features with respect to plant requirements and wine production. The physics of our approach are defined by the use of soil–plant–atmosphere simulation models, applying physically based equations to describe the soil hydrological processes and solve soil–plant water status. This study (part of the ZOVISA project) was conducted on a farm devoted to production of high-quality wines (Aglianico DOC), located in southern Italy (Campania region, Mirabella Eclano, AV). The soil spatial distribution was obtained after standard soil survey informed by geophysical survey. Two homogeneous zones (HZs) were identified; in each one a physically based model was applied to solve the soil water balance and estimate the soil functional behaviour (crop water stress index, CWSI) defining the functional homogeneous zones (fHZs). For the second process, experimental plots were established and monitored for investigating soil–plant water status, crop development (biometric and physiological parameters) and daily climate variables (temperature, solar radiation, rainfall, wind). The effects of crop water status on crop response over must and wine quality were then evaluated in the fHZs. This was performed by comparing crop water stress with (i) crop physiological measurement (leaf gas exchange, chlorophyll a fluorescence, leaf water potential, chlorophyll content, leaf area index (LAI) measurement), (ii) grape bunches measurements (berry weight, sugar content, titratable acidity, etc.) and (iii) wine quality (aromatic response). This experiment proved the usefulness of the physically based approach, also in the case of mapping viticulture microzoning

    Implementation of Whole-Body MRI (MY-RADS) within the OPTIMUM/MUKnine multi-centre clinical trial for patients with myeloma.

    BACKGROUND: Whole-body (WB) MRI, which includes diffusion-weighted imaging (DWI) and T1-w Dixon, permits sensitive detection of marrow disease in addition to qualitative and quantitative measurements of disease and response to treatment of bone marrow. We report on the first study to embed standardised WB-MRI within a prospective, multi-centre myeloma clinical trial (IMAGIMM trial, sub-study of OPTIMUM/MUKnine) to explore the use of WB-MRI to detect minimal residual disease after treatment. METHODS: The standardised MY-RADS WB-MRI protocol was set up on a local 1.5 T scanner. An imaging manual describing the MR protocol, quality assurance/control procedures and data transfer was produced and provided to sites. For non-identical scanners (different vendor or magnet strength), site visits from our physics team were organised to support protocol optimisation. The site qualification process included review of phantom and volunteer data acquired at each site and a teleconference to brief the multidisciplinary team. Image quality of initial patients at each site was assessed. RESULTS: WB-MRI was successfully set up at 12 UK sites involving 3 vendor systems and two field strengths. Four main protocols (1.5 T Siemens, 3 T Siemens, 1.5 T Philips and 3 T GE scanners) were generated. Scanner limitations (hardware and software) and scanning time constraint required protocol modifications for 4 sites. Nevertheless, shared methodology and imaging protocols enabled other centres to obtain images suitable for qualitative and quantitative analysis. CONCLUSIONS: Standardised WB-MRI protocols can be implemented and supported in prospective multi-centre clinical trials.     A Novel Labeling Approach Identifies Three Stability Levels of Acetylcholine Receptors in the Mouse Neuromuscular Junction In Vivo

    The turnover of acetylcholine receptors at the neuromuscular junction is regulated in an activity-dependent manner. Upon denervation and under various other pathological conditions, receptor half-life is decreased., in our setup the same animals are used throughout the whole measurement period, thereby permitting a dramatic reduction of animal numbers at increased data quality. We identified three stability levels of acetylcholine receptors depending on the presence or absence of innervation: one pool of receptors with a long half-life of ∼13 days, a second with an intermediate half-life of ∼8 days, and a third with a short half-life of ∼1 day. Data were highly reproducible from animal to animal and followed simple exponential terms. The principal outcomes of these measurements were reproduced by an optical pulse-labeling assay introduced recently.A novel assay to determine kinetics of acetylcholine receptor turnover with small animal numbers is presented. Our data show that nerve activity acts on muscle acetylcholine receptor stability by at least two different means, one shifting receptor lifetime from short to intermediate and another, which further increases receptor stability to a long lifetime.     Influence of mycotoxin zearalenone and its derivatives (alpha and beta zearalenol) on apoptosis and proliferation of cultured granulosa cells from equine ovaries

    BACKGROUND: The mycotoxin zearalenone (ZEA) and its derivatives, alpha and beta-zearalenol (alpha and beta-ZOL), synthesized by genera Fusarium, often occur as contaminants in cereal grains and animal feeds. The importance of ZEA on reproductive disorders is well known in domestic animals species, particularly in swine and cattle. In the horse, limited data are available to date on the influence of dietary exposure to ZEA on reproductive health and on its in vitro effects on reproductive cells. The aim of this study was to evaluate the effects of ZEA and its derivatives, alpha and beta-ZOL, on granulosa cells (GCs) from the ovaries of cycling mares. METHODS: The cell proliferation was evaluated by using the 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide (MTT) test after 3 days exposure at different concentrations of ZEA and its derivatives (from 1 × 10-7 to 0.1 microM). The apoptosis induction was evaluated after 1 day exposure, by DNA analysis using flow cytometry. RESULTS: An increase in cell proliferation with respect to the control was observed in the presence of ZEA at 1 × 10-3 and 1 × 10-4 microM and apoptosis was induced by all mycotoxins at different concentrations. CONCLUSION: The simultaneous presence of apoptosis and proliferation in GC cultures treated with zearalenones could indicate that these mycotoxins could be effective in inducing follicular atresia. These effects of zearalenones may result from both direct interaction with oestrogen-receptors as well as interaction with the enzymes 3alpha (beta)-hydroxysteroid dehydrogenase (HSD), involved in the synthesis and metabolism of endogenous steroid hormones. These cellular disturbances, described for the first time in equine GCs cultured in vitro, could be hypothesized as referred to reproductive failures of unknown ethiology in the mare

    Image quality in whole-body MRI using the MY-RADS protocol in a prospective multi-centre multiple myeloma study

    Background The Myeloma Response Assessment and Diagnosis System (MY-RADS) guidelines establish a standardised acquisition and analysis pipeline for whole-body MRI (WB-MRI) in patients with myeloma. This is the first study to assess image quality in a multi-centre prospective trial using MY-RADS. Methods The cohort consisted of 121 examinations acquired across ten sites with a range of prior WB-MRI experience, three scanner manufacturers and two field strengths. Image quality was evaluated qualitatively by a radiologist and quantitatively using a semi-automated pipeline to quantify common artefacts and image quality issues. The intra- and inter-rater repeatability of qualitative and quantitative scoring was also assessed. Results Qualitative radiological scoring found that the image quality was generally good, with 94% of examinations rated as good or excellent and only one examination rated as non-diagnostic. There was a significant correlation between radiological and quantitative scoring for most measures, and intra- and inter-rater repeatability were generally good. When the quality of an overall examination was low, this was often due to low quality diffusion-weighted imaging (DWI), where signal to noise ratio (SNR), anterior thoracic signal loss and brain geometric distortion were found as significant predictors of examination quality. Conclusions It is possible to successfully deliver a multi-centre WB-MRI study using the MY-RADS protocol involving scanners with a range of manufacturers, models and field strengths. Quantitative measures of image quality were developed and shown to be significantly correlated with radiological assessment. The SNR of DW images was identified as a significant factor affecting overall examination quality

    Abstract · Notes on Textile production and on Clothing from the medieval city of Siponto · The present work deals with the themes of fabric and clothing archeology in the light of the experimental archeology method. Some structural and material elements coming from archaeological research in the abandoned city of Siponto (Manfredonia, FG), an important port on the Adriatic until the middle of the 13th century, are reconsidered. On the basis of archaeological data, iconographic references, archaeometric results, a vertical frame was created, probably resting on the perimeter of one of the residential buildings of the context and a wool tunic with its accessories, specially reproduced on the model of the finds. The study has thus allowed us to interpret or reinterpret some archaeological traces and to formulate a series of considerations on the structure of the frame and on the development of the relative manufacturing activity.Riassunto · Il presente lavoro affronta i temi dell’archeologia del tessuto e dell’abbigliamento alla luce del metodo dell’Archeologia sperimentale. Vengono riconsiderati alcuni elementi strutturali e materiali venuti in luce dalla ricerca archeologica nella città abbandonata di Siponto (Manfredonia, FG), importante scalo portuale sull’Adriatico fino alla metà del XIII secolo. Sulla base dei dati archeologici, dei riferimenti iconografici, dei risultati archeometrici è stato realizzato un telaio verticale, verosimilmente appoggiato al perimetrale di uno degli edifici abitativi del contesto e una tunica in lana con i suoi accessori, appositamente riprodotti sul modello dei reperti.     Black leaf–clips of a commercial fluorometer increased leaf temperature during dark adaptation under high solar radiation

    The use of black leaf-clips for dark adaptation under high solar radiation conditions is reported to underestimate the maximum quantum yield of PSII photochemistry (Fv/Fm) measured by the continuous-excitation fluorometer Pocket PEA. The decrease in Fv/Fm was due to a rise in minimum fluorescence emission (Fo), probably resulting from increased leaf temperature (Tl). In field-grown tomato and pepper, fluorescence parameters and Tl in the region covered by the black leaf clip were measured in clipped leaves exposed to solar radiation during dark adaptation (clipped-only leaves) and in clipped leaves protected from solar radiation by aluminium foil (shrouded clipped leaves). Results confirmed significant Fv/Fm underestimates in clipped-only leaves primarily due to increased Fo. In one tomato experiment, Tl increased from 30 to 44.5°C in clipped-only leaves, with a negligible rise in shrouded clipped leaves. In two respective pepper experiments, Tl in clipped-only leaves increased from 27 to 36.2°C and 33 to 40.9°C. Based on the results of this study, a clip-effect parameter (PCE) on fluorescence emission is proposed as the difference for Fv/Fm (or –Fo/Fm) between shrouded clipped leaves and clipped-only leaves, which resulted to be 0.706 for tomato, and 0.241 and 0.358 for the two pepper experiments
